Description: | Abstract: Gregarines constitute a large group of endobiontic apicomplexans (Alveolata, Eukaryota), inhabiting different compartments of invertebrate hosts. Despite years of research on gregarines, their relationship, function and interaction with the host and their own evolutionary origin remain poorly understood. In this study, we concentrated on the interaction between eugregarines infecting darkling beetles (Tenebrionidae, Coleoptera) collected from several locations in the Atacama Desert and adjacent areas. Working with morphological and molecular methods we were able to isolate and describe two putative new gregarine species: Atacamagregarina psectrascelii sp. nov. from Salar de Huasco and Atacamagregarina sp. nov. from Quebrada Aroma. Phylogenetic analyses of the 18S rDNA gene of Atacamagregarina sp. nov. revealed a close relationship to Atacamagregarina paposa, infecting the same beetle genus. Atacamagregarina psectrascelii sp. nov. branches in a clade together with species belonging to the genus of Xiphocehalus and Stylocephalus, infecting darkling beetles in North America. One other important finding was that many gregarine cells, found intact in the guts of the hosts, had no nucleus any more. This led us to the assumption that not all cells are viable and DNA cannot be obtained of every cell. Prospective studies will focus on single-cell genomic approaches to obtain greater and more reliable phylogenetic resolution and the possibility to functionally characterize the interaction between eugregarines and their hosts. |
