Title:Main Title: Studies on gregarines inhabiting darkling beetles of the Atacama Desert
Description:Abstract: Gregarines are endobiontic apicomplexans (Alveolata, Eukaryota), found within terrestrial and aquatic invertebrates. Most of them are assumed to be host specific and therefore of special interest for host-endobiont co-evolution and population studies. Gregarines inhabiting darkling beetles of the Atacama Desert are particular interesting for these studies, since the molecular clock of their hosts, as well as the geological origins and tectonic formatting of their habitat been well described. In this study we focused on the establishment of a replicable and universal method for the isolation and amplification of rRNA from gregarines, in preparation for further investigations on the evolutionary processes of the above mentioned gregarines. For this purpose we established Tenebrio molitor larva as a model organism as it inherits at least three different gregarine species, Gregarina polymorpha, G. steinii and G. cuneata. Several different lysis, molecular biological and staining protocols and methods were tested to optimize the yield of information from a single gregarine cell. Several methodical problem sources were identified and eliminated, resulting in a preliminary protocol. Applying this new methods we were able to confirm the SSU rRNA sequences of Atacamagregarina paposa and Atacamagregarina psectrascelii. Further, a gregarine species isolated from Psectrascelis sp. could be sequenced as well. A probably new gregarine species was found in Tenebrio molitor larva and was also be sequenced regarding its LSU rRNA gene. In addition we describe the infection rates of twenty Beetles species, sampled in the Atacama Desert. Further studies will use the newly established method as a standard for isolating and sequencing gregarines, finally enabling us to study evolutionary processes within the complex host-endobiont system of beetles and gregarines from the Atacama Desert.
