Title: | Main Title: Molecular and morphological investigation on protist diversity in the phyllosphere of cacti in Northern Chile |
Description: | Abstract: Due to the extreme conditions in the Atacama Desert, different endemic cacti have evolved through speciation. As each plant forms its own microbiome, those conditions will also have an influence on it, favouring co-evolution. This study focuses on the protist inhabitants of these biomes. Protists are eukaryotic unicellular organisms and the degree of their endemicity is still unresolved. Three different cacti species (Eulychnia sp., Copiapoa sp., Oreocereus sp.) each originating from different locations, were sampled. The general species composition of protists associated with the sampled cacti was investigated by using standardized conditions. In addition, isolated protists were morphological and molecular analysed. As a result, no isolated protist species was found on more than one cactus species and its surrounding microhabitat. Isolated protists were obtained by using the liquid aliquot method and four different phylogenetic trees belonging to colpodid ciliates, cercozoans, bodonids and bicosoecids were created with 18 obtained protist strains. The investigated species composition showed that protist communities associated with cacti may differ depending on the position on the plant. The results of this study indicate the existence of endemic protist species in the Atacama Desert as well as ubiquitous distributed protists. In order to understand the high diversity of protists and to clarify how many endemic species really exist, more samples need to be collected and analysed. The comparison of similar extreme habitats (e.g. Namib Desert) could be of particular interest, as they might be exposed to similar selection pressures. |
