Title:Main Title: Diversity and phylogeny of percolomonads based on newly discovered species from hypersaline and marine waters
Description:Abstract: Percolomonads are common marine, freshwater and hypersaline tetraflagellated organisms. However, up to now all observations have been assigned to only one single species, Percolomonas cosmopolitus, which has been well characterized regarding its morphology. Current phylogenetic trees of eukaryotes comprise only two uncertain genotypes of this important family. Here, we investigated the general morphology, salinity tolerance and 18S rDNA gene based phylogeny of seven percolomonad cultures. We describe three new genera and five novel species: Barbelia atlantica, B. abyssalis, Lula jakobsenorum, Nakurumonas serrata and Percolomonas doradora. Based on large phylogenetic distances and their distinct morphology, we erected two novel families within the percolomonad clade. The new species show features typical for percolomonads, one long flagellum for skidding, three shorter flagella of equal length and a ventral feeding groove. The new species comprise organisms living in athalassic saline, marine and hypersaline waters ranging between 10 and 150 PSU. Based on these novel taxa the taxonomy and phylogeny of Percolatea was extended and revised. Based on the different association of GenBank sequences of “Percolomonas” we reassigned their taxonomic position.
